Cross Protection Experiment in Mice Immunization with Actinobacillus pleuropneumoniae serotype 7 Genomic Expression Library

چکیده
Actinobacillus (A.) pleuropneumoniae is a causative agent of porcine pleuropneumonia, which has been reported in most countries. A genomic expression library of A. pleuropneumonia serotype 7 was constructed to identify potential vaccine candidates. Plasmid DNA from the sub-libraries (L1 to L10) was used to immunize BALB/c mice. Then, 2 weeks after the final immunization, the groups of mice were challenged with A. pleuropneumonia serotype 1 (1×109 cfu). Mice immunized with one of the sublibraries (L3) had a higher level of antibodies and IFN-γ production than others. This sub-library (L3) resulted in a significant reduction in the amount of A. pleuropneumoniae recovered from mouse lungs and the survival rate of the L3 group was the most highest. Our study demonstrates that a genome expression library of A. pleuropneumoniae serotype 7 offers a novel approach for screening possible cross-serotype protection vaccine candidates against A. pleuropneumoniae serotype 1 infection.

The Expression of apxIVA Gene of Actinobacillus pleuropneumoniae Serotype 5 and Establishment of an Indirect ELISA for Distinguishing Infection and Immunization
Actinobacillus pleuropneumoniae is the etiological agent of Porcine pleuropneumonia, which causes severe losses in pig farming. To establish an serological method to detect A. pleuropneumoniae infection, bioinformatics method was utilized to analyzed the sequence of apxIVA gene of A. pleuropneumoniae serotype 5 completed by our lab, a 1152 bp fragment of the N'-terminal of apxIVA gene was ampli...
متن کاملEnhancement of protective immune responses by oral vaccination with Saccharomyces cerevisiae expressing recombinant Actinobacillus pleuropneumoniae ApxIA or ApxIIA in mice
We previously induced protective immune response by oral immunization with yeast expressing the ApxIIA antigen. The ApxI antigen is also an important factor in the protection against Actinobacillus pleuropneumoniae serotype 5 infection; therefore, the protective immunity in mice following oral immunization with Saccharomyces cerevisiae expressing either ApxIA (group C) or ApxIIA (group D) alone...
متن کاملDNA vaccine encoding type IV pilin of Actinobacillus pleuropneumoniae induces strong immune response but confers limited protective efficacy against serotype 2 challenge.
Actinobacillus pleuropneumoniae is a gram-negative bacterial pathogen that causes swine pleuropneumonia, a highly contagious and often fatal disease that occurs worldwide. Our previous study showed that DNA vaccines encoding Apx exotoxin structural proteins ApxIA and/or ApxIIA, are a promising novel approach for immunization against the lethal challenge of A. pleuropneumoniae serotype 1. Vaccin...
متن کاملA trivalent Apx-fusion protein delivered by E. coli outer membrane vesicles induce protection against Actinobacillus pleuropneumoniae of serotype 1 and 7 challenge in a murine model
Actinobacillus pleuropneumoniae (APP) causes serious economic losses in the swine industry, and is the etiologic agent of porcine pleuropneumonia. In this study we have engineered a trivalent Apx fusion protein enclosed in outer membrane vesicles (Apxr-OMV) and studied its immunoprotective efficacy against APP serotypes 1 and 7 challenge in mice. The results showed that the IgG levels in the Ap...
متن کاملVaccination and protection of pigs against pleuropneumonia with a vaccine strain of Actinobacillus pleuropneumoniae produced by site-specific mutagenesis of the ApxII operon.
The production of toxin (Apx)-neutralizing antibodies during infection plays a major role in the induction of protective immunity to Actinobacillus pleuropneumoniae reinfection. In the present study, the gene encoding the ApxII-activating protein, apxIIC, was insertionally inactivated on the chromosome of a serovar 7 strain, HS93. Expression of the structural toxin, ApxIIA, and of the two genes...
